Botulinum neurotoxin is both poison and medicine: botulinum therapy and iatrogenic botulism

چکیده
Botulinum neurotoxin is the most potent poison known worldwide. Over past two centuries, its structure and functions have been studied in detail, obtained pure form. Knowledge of mechanisms action botulinum has determined directions possible use: negative, as a biological (toxin) weapon, positive, an agent that eliminates pathological muscle hypertonicity neuropathic pain can be used for aesthetic purposes (beauty injections).
The lecture presents chronological development both provides detailed analysis various uses medical cosmetic treatment. Particular attention paid to side effects therapy ways overcome them.

Temporal characteristics of botulinum neurotoxin therapy.
Botulinum neurotoxin is a pharmaceutical treatment used for an increasing number of neurological and non-neurological indications, symptoms and diseases. Despite the wealth of clinical reports that involve the timing of the therapeutic effects of this toxin, few studies have attempted to integrate these data into unified models. Secondary reactions have also been examined including the developm...
متن کاملOn Botulinum Neurotoxin Variability
The rapidly growing number of botulinum neurotoxin sequences poses the problem of the possible evolutionary significance of the variability of these superpotent neurotoxins for toxin-producing Clostridium species. To progress in the understanding of this remarkable phenomenon, we suggest that researchers should (i) abandon an anthropocentric view of these neurotoxins as human botulism-causing a...
متن کاملBotulinum neurotoxin: a deadly protease with applications to human medicine
Botulinum neurotoxins (BoNTs) are some of the most potent biological toxins to humans. They are synthesized by the gram-positive, spore-forming bacterium Clostridium botulinum. BoNT is secreted from the bacterium as a ~150 kDa polypeptide which is cleaved by bacterial or host proteases into a ~50 kDa light chain and a ~100 kDa heavy chain disulfide-linked protein. The light chain of the toxin c...
متن کاملBotulinum neurotoxin: the ugly duckling.
This review presents a brief account of the most significant biological effects and clinical applications of botulinum neurotoxins, in a way comprehensive even for casual readers who are not familiar with the subject. The most toxic known substances in botulinum neurotoxins are polypeptides naturally synthesized by bacteria of the genus Clostridium. These polypeptides inhibit acetylcholine rele...

In this issue of Applied and Environmental Microbiology (AEM), we publish a paper by Marite Bradshaw, William H. Tepp, Regina C. M. Whitemarsh, Sabine Pellett, and Eric A. Johnson from the Department of Bacteriology, University of Wisconsin— Madison (UW—M), reporting on the purification and characterization of subtype A4 botulinum neurotoxin (1). The authors describe the purification of recombi...
